How exactly do we tweak?

I posted another message asking about tweaks. What i want to please know, is how exactly do we do this. When i open the files that are supposed to be tweaked, I cant delete and rewrite any of the data thats already there.
Anyone nice enough to explain to me how?

Don't open it, right click to edit it, and it opens, editable, in notepad.
